VIP (Vasoactive Intestinal Peptide)

VIP is a 28-amino acid neuropeptide originally isolated from the small intestine but now known to be widely distributed throughout the central and peripheral nervous systems, immune system, and gastrointestinal tract. It belongs to the glucagon/secretin superfamily and acts through two G-protein coupled receptors (VPAC1 and VPAC2). VIP has generated significant research interest due to its broad range of physiological roles spanning immune regulation, neuroprotection, and circadian biology.

Mechanism of Action

VIP activates VPAC1 and VPAC2 receptors, both of which are coupled to adenylyl cyclase and increase intracellular cAMP. Through VPAC1, VIP exerts potent anti-inflammatory effects by inhibiting NF-kB activation, reducing pro-inflammatory cytokine production (TNF-alpha, IL-6, IL-12), and promoting regulatory T-cell generation. Through VPAC2, it modulates circadian rhythms via the suprachiasmatic nucleus, regulates smooth muscle relaxation, and supports bronchodilation. VIP also promotes neuronal survival through BDNF upregulation and inhibits apoptosis in multiple cell types.
