SS-31 (Elamipretide)

SS-31, also known as Elamipretide and MTP-131, is a synthetic tetrapeptide (D-Arg-Dmt-Lys-Phe-NH2) that selectively targets and concentrates within the inner mitochondrial membrane. It is the first compound of its kind - a mitochondria-targeted peptide that directly stabilizes the electron transport chain and restores bioenergetic function. SS-31 has been the subject of extensive preclinical and clinical research across multiple disease models linked to mitochondrial dysfunction.

Mechanism of Action

SS-31 selectively binds to cardiolipin, a phospholipid unique to the inner mitochondrial membrane that is essential for the structural organization of electron transport chain complexes. By stabilizing cardiolipin interactions, SS-31 optimizes electron transport chain efficiency, reduces electron leak, and decreases the production of reactive oxygen species (ROS) at their primary source. This is mechanistically distinct from conventional antioxidants, which scavenge ROS after they are produced. SS-31 concentrates in mitochondria at 1000-5000x its extracellular concentration due to its Szeto-Schiller peptide structure.
